Teza

In recent years, the green microalgae Dunaliella salina, an important biotechnological object, is in the area of great interest of researchers [1]. Dunaliella biomass is used for the obtaining of numerous bioactive substances with antioxidant properties, such as: lipids (unsaturated fatty acids), carotenoids (β- carotene, zeaxanthine, luteine), vitamins (α - tocopherol) et. al. It is known that carotenoids preponderantly prevent the formation of peroxyl radicals and forms of singlet molecular oxygen, as well as suppress the processes of peroxidical oxidation of lipids [2]. Besides of β- carotene other forms of carotenoid from dunaliella biomass – zeaxanthine and luteine have the antioxidant properties too [3]. Recently, the positive effect of some coordination compounds of Cr(III), Fe(III) and Co(II) on Dunaliella salina productivity and carotenoidgenesis was established by collaborators of research laboratory „Phycobiotehnology” [1]. The aim of investigation: the obtaining of Dunaliella salina biomass with high carotenoids content. The object of study: green microalgae Dunaliella salina CNM–AV-01, deposited in the National Collection of Non-Pathogen Microorganisms of Institute of Microbiology and Biotehnology of ASM. Dunaliella was cultivate don the nutritive medium Ben – Amotz [4]. The coordination compounds of Zn(II) with organic acids and imidazole as ligands, supplemented to the nutritive medium in the concentrations of 0,1-30 mg/L, have been used as stimulators of carotenoidgenesis. The carotenoids content was determined by the method of Dere [5]. According to the results of investigation, the coordination compounds of Zn(II) with organic acids and imidazole can be used as stimulators of carotenoidgenesis in the dunaliella biomass. The optimal concentration has been established (10 mg/L). The complex of Zn with adipic acid and imidazole has had the maximal effect on the carotenoids content (an increasing by 55 % compared to the control). This positive effect can be caused by the presence of zinc in the active center of several important enzymes of carotenoids biosynthesis. Such, ions of Zn2+ are included in the composition of enzyme DMAPP-isomerase catalysing one of the essential reaction of izoprenoid way – the conversion of IPPP to DMAPP. Another important enzyme – farnesyltranspherase taking part in the reaction of synthesis of geranyl geranylpyrophosphate contain in the active center ions of zinc also. [6]. The biomass of Dunaliella salina with high content of carotenoids can be recommended for the obtaining of bioadditives with the following utilization in treatment and profilaxy of cancer, infectious and cardio-vascular diseases. The advantage of these bioadditives is in the presence of 9- cis isomers of β – carotene.
