The present work reports on a study of plant regeneration carried out with callus from the leaf blades and petioles on femal plants of Actinidia cinensis. In order to optimize the culture medium for callus induction and following regeneration of plantlets were tested 7 types of medium, with different combinations of growth regulators. Best result of callus induction were obtained on MS added with 0,5mg/l BAP and 0,25mg/l ANA, B5 with 0,04 mg/l BAP and 0,2 mg/l IBA, S 2,5 with 1mg/l zeatin and 0,02mg/l ANA. The frequency of callus derived from petiole (81,48%) was higher than those from the leaf blades (69,46%). The influence of the composition of culture medium and types of explants on callus induction was established. The medium S2,5 supplemented with 1,0 mg/l BAP and 1,0 mg/l GA3 had been more effective for shoot formation and plant regeneration. For rooting, developed shoots were obtained on MS medium in the presence of BAP and IBA. |
