Background. The cardiotoxicity of doxorubicin, an antineoplastic drug, it is imposed by the development of cardiomyopathy and heart failure. The expression of ET-1 in myocardium under the action of Dx, direct correlates with the degree of cardiac dysfunction, mediated by ETA receptor. Objective of the study. Evaluation in vitro of ET-1 effects on the functional reactivity of the heart in doxorubicin disorder. Material and Methods. Cross-sectional study. 2 groups of white rats (experimental group n=9, control group n=9) were used. In the control group Dx during 2 weeks was administrated (i/p, 4mg/kg in one dose, twice/week), cumulative dose – 16 mg/kg. The ET-1 effects were estimated at its peak action in concentration 10-7 M (mol), reproduced after 30 sec of endothelin stimulation. Results. The functional parameters of isolated heart perfused in physiologic regime under the ET-1 action in group with Dx compared with the control one: cardiac output (CO) – 21,5±1,5 vs 42,7±2,2 ml/min; LV systolic pressure (LVSP) – 95,4±6,6 vs 177,3±8,6 mm Hg; LV end-diastolic pressure of (LVEDP) – 17,7±1,26 vs 6,1±0,55 mm Hg. In condition of volume overload, under the ET1 action: CO – 28,6±2,6 vs 51,3±4,7 ml/min; LVEDP – 24,8±2,44 vs 7,5±0,72 mm Hg, but in condition of resistance overload, the CO – 11,4±1,22 vs 25,4±2,23 ml/min; LVSP – 111,5±10,6 vs 172,8±14,7 mm Hg. Conclusion. 1) Under the ET-1 action on the isolated heart perfused in physiologic regime in the group with Dx was shown negative inotropic effect. 2) At the volume and resistance overload test under the ET-1 action were demonstrated the impairment of diastolic function and myocardial contractile capacity. |
