Болезнь Паркинсона (БП) – одно из самых распространенных нейродегенеративных заболеваний, после болезни Альцгеймера, приводящее к двигательным нарушениям. В развитых странах БП поражает около 2% населения после 65 лет. Сипмтомами заболевания являются: акинезия, ригидность лицевых и скелетных мышц и тремор покоя. Характерным признаком заболевания является наличие телец Леви, образующихся в результате скопления в цитоплазме белка альфа-синуклеина (SNCA), относящегося к семейству амилоидных белков, а также дегенерация дофаминергических нейронов в переднем отделе черной субстанции. Перспективной группой веществ, препятствующих образованию амилоидных фибрилл, являются производные имидазола. Соединение – 5-фосфат-рибозил-аминоимидазол (АИР) лежит в основе полимера, образующегося в клетках дрожжей Saccharomyces сerevisiae при некоторых мутациях в пути биосинтеза аденина и придающего колониям дрожжевых клеток характерный красный цвет, поэтому этот полимер получил название «красный пигмент». В качестве модельного объекта в нашей работе использовалась плодовая мушка Drosophila melanogaster с экспрессией гена альфа-синуклеина человека дикого типа и двух его мутантных форм: А30Р и А53Т. Мы исследовали уровень дегенерации дофаминергических и холинергических нейронов в клетках мозга мух, а также оценили уровень отложений растворимой и нерастворимой формы альфа-синуклеина, при кормлении мух красными и белыми дрожжами. В результате мы показали, что количество альфа-синуклеина как дикого типа, так и мутантных форм снижалось при кормлении мух красными дрожжами, а также значительно был снижен уровень дегенерации дофаминергических нейронов. Количество холинергических нейронов в течение эксперимента у мух, содержащихся на белых или красных дрожжах не отличалось. Мы считаем, что снижение уровня нейродегенерации, а также общего уровня белка альфа-синуклеина происходит за счет действия 5- фосфат-рибозил-аминоимидазола, как компонента красных дрожжей.
Parkinson disease (PD) is the most frequent movement disorder and the second most common neurodegenerative disorder after Alzheimer’s disease. In developed countries, PD affects 1–2% of people over 65 years of age. Symptoms of PD include akinesia, rigidity of facial and skeletal muscles, resting tremor. Presence of Lewy bodies in the brain cells and degeneration of dopaminergic neurons in the substantianigra pars compacta are the characteristic features of the disease. Accumulation of the alpha-synuclein protein (SNCA), which belong to the family of amyloid proteins, in the cytoplasm leads to the formation of the Lewy bodies. A group of substances, which are derivatives of imidazole, inhibits formation of amyloid fibrils and has a potential for development into anti-PD drugs.. The 5-phosphate-ribosyl-aminoimidazole compound forms a polymer, which is formed in the Saccharomyces cerevisiae yeast cells with specific mutations in the pathway of adenine biosynthesis. It gives the colony of yeast cells a characteristic red color, therefore this polymer is called "red pigment". As a model subject, we used in our experiments a fruit fly Drosophila melanogaster expressing three variants of human alpha-synuclein gene: wild-type or its two mutant forms: A30P and A53T. We investigated the level of degeneration of dopaminergic and cholinergic neurons in cells of the fly brain, as well as assessed the level of sedimentation of the soluble and insoluble forms of alpha-sinuclein in flies fed with red and white yeast. As a result, we have demonstrated that in flies fed with red yeast the amount of alpha-synuclein of both wild type and mutant forms decreased, and the level of degeneration of dopaminergic neurons significantly reduced compared to flies fed with white yeast. The number of cholinergic neurons during the experiment in flies fed with white or red yeast was similar. We believe that the decrease in the level of neurodegeneration, and in the overall level of the alpha-sinuclein protein, is due to the action of 5-phosphate-ribosyl-amino imidazole, as a component of red yeast.
