Background. Gallstones are a common condition with multiple etiologies and risk factors, such as: female gender, obesity, age, high-calorie diet, etc. In about 80% of cases, cholesterol stones that crystallize and settle in the bladder and bile ducts are determined. Objective of the study. Study of choleretic and cholagogue drugs (CCD), their biopharmaceutical and pharmacokinetic evaluation. Material and Methods. The main national and international electronic sources of information were selected: Scopus and PubMed. Results. 85% of the more than 50 analyzed publications that have been conducted in the case of gallstones revealed that the drug treatment is effective in prophylactic terms, especially in people at risk. Cholesterol, the most common component of gallstones, was solubilized in the bile by bile acids and amino acids loaded into liposomes. Drug therapy encounters difficulties due to the fact that the maximum share of CCD is part of biopharmaceutical class 2, has high lipophilicity, octanol/water partition coefficient from 3 to 4 and low solubility, average logS of -4.8 mol/L, requiring advanced complexation technologies with hydrotropic agents. A promising alternative is the water-soluble derivatives of hydroxycinnamic acids: ferulic acid and cinnarine, which have the advantage of multiple antioxidant, anti-inflammatory, antibacterial, lipolipedemic and metabolic actions. Conclusion. At present, in the field of biopharmaceuticals, the methods of incorporating CCD in β-cyclodextrin and other polymers, in combination with sodium salicylate, aiming the optimization of their medicinal effect, are being studied extensive. |
