Известно, что монокулярная депривация влияет на механизмы нейронной пластичности, лежащие в основе зрительно – двигательных реакций. Восстановление зрительной функции после монокулярной депривации протекает медленно и не в полном объеме. Целью данной работы является поиск новых подходов, приводящих к восстановлению морфологии дендритов командных нейронов, принимающих зрительную информацию и преобразующих ее в двигательные акты. Было показано, что инкубация ганглия моллюска прудовика Lymnaea stagnalis в присутствие различных концентраций тетрапептида Thr-Ser-Lys- Tyr (TSKY), выделенного из мозга сусликов Spermophillus undulatus в состоянии спячки, достоверно увеличивала общее количество живых нейронов в культуре. Для определения эффекта данного пептида на восстановление структуры и функции нейронов, было исследовано его влияние на морфологию дендритов парных маутнеровских нейронов (МН) рыб Carassius auratus. Мы разработали метод монокулярной депривации, суть которого заключается в обратимом закрывании одного глаза мальков 6 мес возраста клеевой маской на срок 1-3 недели. Мозг рыб фиксировали в растворах глутарового альдегида и четырехокиси осмия и пропитывали эпоксидной смолой. Морфологию МН изучали, анализируя фронтальные срезы medulla oblongata с помощью световой микроскопии. Объемные характеристики главных дендритов и их ветвей 1 и 2 порядка определяли, используя методику трехмерной (3D) реконструкции серийных гистологических срезов толщиной 3 мкм. Исследование моторного поведения рыб показало, что функциональная активность нейрона, являющегося мишенью депривации, значительно нарастает, а активность его парного, контрольного нейрона снижается. Аппликации TSKY на МН, проводившиеся после отклеивания глазной маски, обеспечивают ускорение восстановления функциональной активности обоих МН до исходного уровня. Пептид TSKY способен сохранять морфологию МН депривированных рыб, укорачивая стволовые части дендритов МН и не меняя их дистальные отделы. Мы полагаем, что TSKY приводит к стабилизации уровня структурной пластичности дендритов МН, подвергавшихся монокулярной депривации. Monocular deprivation affects mechanisms of neuronal plasticity which are responsible for visual motor behavior. Recovery of visual function from monocular deprivation is slow and incomplete. We investigated a new approach to improve the dendrite morphology of a second-order sensory neurons which receive visual signals and convert them to motor acts. Earlier we studied the survival of cultured neurons from the pond snail Lymnaea stagnalis and demonstrated the effect of addition to the incubation medium of the peptide Thr-Ser-Lys-Tyr (TSKY) isolated earlier from the brain of the ground squirrel Spermophillus undulatus. An increase in cell viability in culture was observed after the treatment of the brain with TSKY at different temperatures. In this study, we show the impact of TSKY on the recovery of neurons following monocular deprivation and investigate the morphology of dendrites of the paired Mauthner neurons (MNs) of goldfish Carassius auratus. We have developed the method of monocular deprivation which is initiated by reversible sealing one eye of the 6- month-old goldfish. The sealing was maintained for 1-3 weeks. The brain of goldfish was fixed in glutaraldehyde and osmium tetroxide solution and embedded in epoxy resin. Cross-sections were obtained, and morphology of MNs was analyzed by light microscopy. The volume characteristics of the primary dendrites and the branches were determined by the three-dimensional (3D) reconstruction of the serial 3-μm histological medulla oblongata sections. The study of goldfish motor behavior revealed that the functional activity of MNs being subjected to monocular deprivation was increased and the activity of control MNs was decreased. Application of TSKY onto MNs location area after reopening the eye lead to dramatic acceleration in the recovery of the functional activity of these paired neurons and returned it to the pre-operative level. This tetrapeptide was able to stabilize the structure of MNs by shortening the basal parts of the dendrites in the brain of the goldfish after operation. The distal parts of dendrites remain unchanged. We believe that TSKY leads to stabilization of structural plasticity in dendrites of MNs following monocular deprivation.
