В последние годы углеродные наноматериалы находят все большее применение в различных отраслях промышленности. Фуллерены, нанотрубки и наноалмазы используют в оптике, электронике, косметологии и фармацевтике. Однако биологические эффекты наночастиц исследованы недостаточно для того, чтобы делать какие – либо выводы о вреде или пользе для окружающей среды и здоровья человека. В то же время перспективы их использования в медицине огромны. Например, такие свойства фуллеренов, как их антиоксидантная активность, способность препятствовать агрегации амилоидных белков делают их перспективными для нейробиологии, повышенная токсичность в отношении опухолевых клеток – в онкологии. Кроме того, фуллерены можно использовать в качестве векторов для доставки лекарственных препаратов, для увеличения контрастности МРТ и др. Эффекты фуллеренов и их растворимых производных – фуллеренолов зависят от многих факторов, например, для функционализации фуллеренов, обладающих слабой биодоступностью,.используют различные растворители, в том числе, токсичные. При недостаточной очистке в процессе синтеза мы получаем в той или иной степени токсичный препарат. Кроме того, есть данные о том, что биологические свойства наночастиц зависят от их способности образовывать кластеры и от величины этих кластеров. То есть проблема приготовления растворов наночастиц заданного состава и с регулируемыми свойствами является одной из основных при их практическом применении. Целью нашей работы было исследование биологических эффектов фуллеренолов С60, С70 и С120, в том числе, приготовленных разными способами. Мы проанализировали токсичность исследуемых препаратов in vitro (на культурах различных клеток) и in vivo на Drosophila melanogaster. Показано, что все препараты обладают низкой токсичностью in vivo, однако на клеточном уровне этот показатель зависел и от препарата, и от способа его приготовления, и от используемой клеточной линии. Следующей нашей задачей был анализ нейропротекторных свойств фуллеренолов. Все исследованные нами фуллеренолы проходили гематоэнцефалический барьер, не обладали нейротоксичностью, то есть не индуцировали нейродегенерацию и имели нейропротекторные свойства. На различных моделях нейродегенеративных заболеваний на Drosophila melanogaster мы наблюдали снижение гибели нейронов при добавлении фуллеренолов в корм животных, уменьшение количества нерастворимых амилоидных белков в мозге мух и другие положительные эффекты. In recent years, carbon nanomaterials have been increasingly used in various industries. Fullerenes, nanotubes and nanodiamonds have been used in electronics and optics, as well as in cosmetology and pharmaceutics. However, the biological effects of nanoparticles have not been explored sufficiently to draw any certain conclusions about a harm or benefits to the environment and the human health. At the same time, opportunities for their use in medicine are enormous. For example, certain properties of fullerenes, such as antioxidant activity and ability to inhibit the aggregation of amyloid proteins make them promising substances for application in neurobiology, while increased toxicity towards tumor cells – in oncology. In addition, fullerenes can be used as drug vectors, to increase the contrast of MRT, etc. Numerous studies have shown that the effects of fullerenes and their soluble derivatives, fullerenols, depend on many factors. One of the main factors, in our opinion, might be a method of preparation. It is known that fullerenes are insoluble in water and have weak bioavailability. Functionalization of fullerenes requires the use of various solvents, including the toxic ones. Inappropriate purification schemes may result in some toxicity of the final drug preparation. In addition, there is evidence that biological properties of nanoparticles depend on their ability to form clusters and on the size of these clusters. Hence, the problem of preparing solutions of nanoparticles of a given composition and specific desired properties is one of the main problems in their practical application. The goal of our work was to study biological effects of fullerenols C60, C70 and C120, including those prepared in various ways. We analyzed toxicity of the test preparations in vitro using cultures of different cells and in vivo using Drosophila melanogaster model. It has been shown that all drugs have low toxicity in vivo, but at the cellular level the toxicity index depends on the preparation itself, on the method of its synthesis and purification, and on the cell line used. Our next task was to analyze the neuroprotective properties of fullerenols. In our experiments, all fullerenols have been able to pass the blood-brain barrier and have not possessed neurotoxicity. Consequently, they have not induced neurodegeneration but instead had neuroprotective properties. In various models of neurodegenerative diseases using Drosophila melanogaster fed with the addition of fullerenols, we have observed a decrease in neuronal death, a decrease in the amount of insoluble amyloid proteins in the brains of flies, and other positive physiological effects.
