Глюкагоноподобный пептид-1 (ГПП-1) – гормон, секретируемый L-клетками подвздошной кишки в кровь и вызывающий глюкозозависимую секрецию инсулина и, как следствие, нормализацию уровня гликемии. В нашей лаборатории, а также в работах других авторов [Perez-Tilve et al., 2010], была показана парадоксальная способность миметика ГПП-1, эксенатида, повышать уровень глюкозы в крови у крыс. По некоторым данным это обусловлено активацией симпатической нервной системы воздействием миметика. Цель работы – определить механизм развития гипергликемии после введения эксенатида. У самок крыс линии Вистар определяли концентрацию глюкозы в крови на 0, 30, 60, 90 и 120 мин от введения миметика ГПП-1. Дозы препаратов и полученные данные были рассчитаны на 100 г массы тела. Различия считали значимыми при p<0.05. После внутримышечного введения эксенатида в дозе 0.5 нмоль происходило развитие гипергликемии, достигавшей максимума (8.2±0.2 мМ) к 30 мин и сохраняющейся на протяжении всего эксперимента. Предварительное (за 30 мин до миметика) внутрибрюшинное введение животным ганглиоблокатора пентамина в дозе 3 мг задерживало развитие эксенатид-стимулированной гипергликемии на 30 мин, что могло быть связано с предотвращением выброса катехоламинов в ответ на введение эксенатида. Для проверки предположения о том, что повышение уровня глюкозы опосредованно действием катехоламинов, за 30 мин до инъекции эксенатида животным внутрибрюшинно были введены неселективные или селективные β-адреноблокаторы. Неселективный β-адреноблокатор пропранолол в дозе 0.5 мг вызвал снижение гипергликемии в течение всего периода наблюдения. Введение селективного β1-адреноблокатора атенолола в дозе 0.2 мг не препятствовало повышению концентрации глюкозы в крови. В тоже время селективный β2-адреноблокатор ICI-118-551, введенный в дозе 0.1 мг, снизил эксенатид- индуцированную гипергликемию через час от начала опыта. Введение адрено– и ганглиоблокаторов без эксенатида не влияло на уровень глюкозы в крови. Таким образом, результаты исследования свидетельствуют о том, что повышение концентрации глюкозы после введения эксенатида опосредовано выбросом катехоламинов и их влиянием на β2-адренорецепторы, что согласуется с данными о β2- адренергической стимуляции процесса гликогенолиза в печени. Glucagon-like peptide-1 (GLP-1) is a hormone secreted by L-cells of the ileum in blood and causing glucosedependent secretion of insulin and, as a consequence, normalization of the level of glycemia. The paradoxical ability of the mimetic GLP-1, exenatide, to raise the level of glucose in the blood was shown in our laboratory, as well as in the works of other authors [Perez-Tilve et al., 2010]. According to some data, this is due to the activation of sympathetic nervous system by the mimetic effect. The aim of the work is to determine the mechanism of development of hyperglycemia after the administration of exenatide. The blood glucose concentration was determined at 0, 30, 60, 90 and 120 minutes from the administration of GLP-1 mimetic to female Wistar rats. Doses and the data were calculated per 100 g of body weight. Differences were considered significant at p<0.05. Hyperglycemia developed after intramuscular administration of exenatide at a dose of 0.5 nmol, it reached a maximum (8.2±0.2 mM) by 30th min and was maintained throughout the experiment. Preliminary (30 min prior to mimetic) intraperitoneal administration of ganglionic blocker pentamine at a dose of 3 mg delayed the development of exenatide-stimulated hyperglycemia in animals for 30 min, this could be due to the prevention of release of catecholamines in response to administration of exenatide. Non-selective or selective β-adrenoblockers were injected intraperitoneally to animals 30 min before the injection of exenatide to test the hypothesis that the increase in glucose level is mediated by the action of catecholamines. Non-selective β-adrenoblocker propranolol at a dose of 0.5 mg caused a decrease in hyperglycemia throughout the observation period. The introduction of selective β1- adrenoblocker atenolol at a dose of 0.2 mg did not prevent the rise of blood glucose. At the same time, the selective β2-adrenoblocker ICI-118-551, administered at a dose of 0.1 mg, reduced exenatide-induced hyperglycemia in an hour from the start of the experiment. The introduction of adrenergic and ganglionic blockers without exenatide did not affect the level of blood glucose. Thus, the results of the study suggest that the increase in the level of glucose after the administration of exenatide is mediated by the release of catecholamines and their effect on β2-adrenoreceptors, which agrees with the data on β2-adrenergic stimulation of the glycogenolysis in the liver.
