Actinomycetes of the genus Streptomyces is a group of Gram-positive bacteria which has rather difficult morphological differentiation with formation of specialized structures such as substrate and aerial mycelium and spores. The basic habitats are the soil and layers of sea and fresh water, besides actinomycetes are found out in air, basically, in the form of spores. Actinomycetes, including the most widespread in the nature genus Streptomyces, are distinguished by a variety of biologically active substances among microorganisms which are factors of development and growth of agricultural animals and plants. These are antibiotics, vitamins, enzymes, lipids, amino acids, phytohormones and others. Nowadays are allocated and described more than 15000 antibiotics. A considerable quantity of these substances is produced by strains, belonging to actinomycetes, mainly, to one of genus – Streptomyces. Despite a variety and a considerable quantity of recipes preparation of media for cultivation of actinomycetes, there are the questions connected with increase of quantity of the received biomass and decrease of its cost, and also increase in the yield of those or others metabolites of investigated producers. In this connection there is a constant search of components which are added to medium for the desired effect. Besides it is necessary to consider concentration of substances in a nutrient medium. Object of study was strain Streptomyces fradiae 19 isolated from soils of the Central Part of Moldova. For antimicrobial properties examination the investigated strain grew up on agar media Capek, Gause and medium R, containing in quality of the basic sources of carbon and nitrogen – starch, corn flour. Antimicrobial activity was defined by using the method of agar blocks. The following testmicroorganisms were used: opportunistic pathogenic microorganisms – E. coli, Ps. aeruginosa, Str. faecalis, St. aureus, Cand. albicans, which were cultivated on medium АGV (composition: pancreatic hydrolyzed of fish flour, sodium chloride, starch, sodium phosphate dibasic, agar, pH – 7,2 – 7,6). At cultivation on the Capek medium, zones of growth inhibition of St. aureus, Cand. albicans and Ps. aeruginosa of 19.0, 35.0 and 10.00 mm accordingly were noticed. At cultivation on the Gause medium, almost full suppression of Cand. albicans growth and zones of growth inhibition of St. aureus and Str. faecalis of 20.0 and 16.0 mm in diameter accordingly were observed . The strain S. fradiae 19, cultivated on medium R, also has shown a high antibacterial activity concerning St. aureus (growth inhibition zones – 18.0 mm), Cand. Albicans (31.0 mm), Ps. aeruginosa (9.5 mm), and Str. faecalis (13.5 mm). Thus, the conducted researches show that metabolites of Streptomyces fradiae 19, isolated from soils of Moldova, have shown the greatest antimicrobial activity at strain cultivation on the Gause medium. |
