Previously we revealed that mRNA levels of VaCPK13 were considerably up-regulated under low temperature stress, mRNA level of VaCDPK20 was significantly up-regulated under cold stress and mRNA level of VaCDPK21was up-regulated under heat and osmotic stress (Dubrovina et al., 2013).We obtained four transgenic cell lines of V. amurensis over expressing the VaCPK13 gene and empty vector to establish control cell lines. The over expression of VaCPK13 gene inhibited the growth of four transgenic cell lines. The data suggested that VaCPK13 gene did not regulate the response of V. amurensis to cold, osmotic and salt stress. We obtained five transgenic cell lines of V. amurensis over expressing the VaCPK20 gene and three transgenic cell lines of Vitis amurensis over expressing the VaCDPK21 gene. Also we transformed A. thaliana with the VaCPK20, VaCDPK21 genes and empty vector to establish control plants in planta. Then, we obtained transgenic A. thaliana plants over expressing VaCPK20 gene and overexpressing VaCDPK21 gene. According to the analysis, over expression of the VaCPK20 gene increased resistance of two V. amurensis cell lines to cold and heat stress and only slightly increased resistance to NaCl treatments. The data suggested that VaCPK20 gene regulated the response of V. amurensis to cold stress and drought stress.The data also suggested that VaCPK21 gene could regulate the response of V. amurensis to salt stress |
