Регуляция функций ЦНС внутренней средой организма может осуществляться через циркум- вентрикулярные органы, в которых гематоэнцефалический барьер теряет свою непроницаемость. В этом плане интерес представляет область area postrema, которая вместе с ядром солитарного тракта и дорзальным моторным ядром блуждающего нерва составляет дорсальный вагусный комплекс. Молекулярные детерминанты изменения барьерных свойств гематоэнцефалического барьера (ГЭБ) головного мозга остаются мало изученными. Изменение свойств ГЭБ может быть обусловлено различием в эпителии сосудов головного мозга клаудинов – белков плотных контактов, определяющих парацеллюлярный транспорт в эпителии и эндотелии. Цель исследования заключалась в изучении экспрессии клаудинов в лобных долях и area postrema головного мозга крыс, отличающихся проницаемостью ГЭБ. Использовали метод иммуногистохимии с анализом изображений в конфокальном лазерном сканирующем микроскопе. В ткани лобных долей головного мозга крысы клаудин-2, обеспечивающий парацеллюлярный транспорт ионов натрия и воды, и клаудин-17, способствующий диффузии анионов, были определены в эндотелии кровеносных сосудов. Об этом свидетельствуют конфигурации меток и их размеров. Наличие сигнала в других клетках нервной системы обнаружено не было. Ко-локализация с окклюдином (белок маркер плотных контактов) свидетельствовала об их включении в структуру плотных контактов. В области area postrema клаудин-2 и -17 не обнаружены. Клаудин-17 детектируется по краю зоны area postrema и за ее пределами. Распределение белков плотных контактов клаудина-2 и -17 в областях с выраженным ГЭБ (лобные доли) и в зонах с его отсутствием (area postrema) имеет региональные отличия. The regulation of the functions of the central nervous system by the internal environment of the body can be carried out through the circum-ventricular organs, in which the blood-brain barrier loses its impermeability. In this respect, the area postrema region, which together with the nucleus of the solitary tract and the dorsal motor nucleus of the vagus nerve, constitutes a dorsal vagal complex. The molecular determinants of the change in the barrier properties of the blood brain barrier (BBB) remain poorly studied. The change in the properties of BBB can be due to the difference in the epithelium of cerebral vessels of the claudins – proteins of tight junction, which determine paracellular transport in the epithelium and endothelium. The aim of the study was to study the expression of claudins in the frontal lobes and area postrema of the brain of rats differing in the permeability of the BBB. The method of immunohistochemistry with image analysis in a confocal laser scanning microscope was used. In the tissue of the frontal lobes of the rat brain, claudin-2, which provides paracellular transport of sodium ions and water, and claudine-17, which promotes the diffusion of anions, were determined in the endothelium of blood vessels. This is evidenced by the configuration of the marks and their sizes. The presence of a signal in other cells of the nervous system was not found. Co-localization with occludin (a marker of tight junction) indicated their inclusion in the structure of tight junction. In area postrema, claudin-2 and -17 were not detected. Claudine-17 is detected along the edge of the area postrema and beyond. The distribution of tight junction proteins claudin-2 and - 17 in areas with BBB (frontal lobes) and in areas with its absence (area postrema) has regional differences.
