Teza

The quantitative tests are often needed in the microbiological studies. Such necessity appears in the investigation of the microbiocenoses, in assessing the quality of nutrient media, in the application necessity of the metered loading doses of microorganisms, in the microbiological evaluation of foods purity, etc. Quantitative counts carried out by the use of microbiological samples inoculation on solid nutrient medium, followed by counting the number of appeared colonies and in the counting of the microbial cells in various suspensions using a counting chamber. The accuracy of calculations strictly depends on the number and multiplicity of dilutions, since the errors associated with the technique of their preparation appear inevitably. The cell counts in squares of counting chambers are impossible, when it is needed to count very dense suspensions because of the repeated calculations of the same cells or because of missed accounted cells. Working with microscope for a long time causes eye fatigue, and leads to a reduction of the researcher’s eyesight. The quantitative counts are always connected with the necessity of documenting and archiving of research results. In order to eliminate these disadvantages the special mechanical and automatic colony counters were invented. These devices make the work much easier and improve its accuracy, but are expensive and their purchase therefore becomes feasible only for specialized laboratories. To overcome these difficulties we have proposed the method of quantitative counts using digital images with detailed description of its application [1, 2]. The use of the digital images for microorganisms counting in the counting chambers shortens the time of working with the microscope and, thus, reduces the harmful effects of stress on the researcher’s eyes, expands the periods of the analysis and allows the researcher to use better his working time.Application of the recorded microbial cells markings allows conducting the counting in suspensions with high microbial cells concentrations, which reduces the analysis error by reducing the number of the suspensions multiplicity dilutions. Improving the analysis accuracy is also achieved due to the possibility of increasing the number of microorganisms cells, counted in the chamber’s squares. Varying color palette allows conducting differentiated elements account in heterogeneous suspensions, containing elements of different quality surveys. The proposed method of accounting colonies in Petri dishes can be used in any microbiological laboratory. The advantage of this method over the other ones is the absence of necessity to use additional, often expensive, equipment. Obtaining digital images makes the method not only more comfortable, more accurate, but also gives it additional quality tool to document the results of work.
