Hemorrhagic shock (HS) results in pronounced increases in circulating concentrations of aspartate (AST), alanine (ALT) aminotransferase, glutamate dehydrogenase (GLDH), IL-1α, IL-6 and TNF-α concentrations associated with histologic alterations of the liver, including midzonal necrosis, hydropic changes, polymorpholeukocyte infiltration. These changes in histologic and clinical chemistry are secondary to tissue hypoxia associated with hypovolemic shock. Administration of Difetur (iNOS inhibitor) after the induction of severe HS significantly attenuated the proinflammatory response both systemically and locally in the liver. Treatment with Difetur prevented neutrophil infiltration into the liver and subsequent liver damage. |
