Cytosolic fumarase as tumor suppressor
It was found that cytosolic fumarase is less involved in the tricarboxylic acid cycle, compared with the mitochondrial one. The study shows exciting connection between primary metabolism (represented by fumarase and fumaric acid) and the DNA damage response. In the absence of fumarase in the cells, its function in DNA repair may be substituted by high concentrations of fumaric acid. It suggests that cytosolic fumarase is not a DNA repair enzyme/protein, but plays an important role in the detection or repair of DNA damage, mainly of DNA double-strand breaks (DSBs). This explains its tumor suppressor activity and the possibility of cancer prevention and treatment. The review reflects the scientific results in this field. |

Studiul relevă legătura dintre metabolismul primar (reprezentat de fumarază şi acidul fumaric) şi răspunsul celular la erorile de la nivelul DNA-ului. La absenţa fumarazei în celule, funcţia sa în repararea DNA-ului poate fi completată de concentraţii ridicate de acid fumaric. Se sugerează faptul că, fumaraza citosolică nu este o enzimă/proteină ce participă direct la repararea DNA-ului, dar joacă un rol important în detectarea sau restabilirea unor erori apărute în structura DNA-ului, îndeosebi a rupturilor bicatenare, fapt ce explică activitatea sa de supresor tumoral, putînd fi implicată în prevenirea şi tratamentul cancerului.
