Структура и функции трансмембранных белков критически зависят от свойств окружающего липидного бислоя. Такие свойства липидного бислоя как толщина (зависит от длины жирных кислот), фазы и текучести (зависит от температуры, степени насыщения жирных кислот, наличия холестерина и сфингомиелина), кривизна (зависит от геометрии головных групп и состава фосфолипидов диацилглицеринов и склонности к образованию ламеллярных или гексагональных структур) влияют на активность сигнальных белков. Если какое-либо химическое вещество внедрено в липидный бислой или наоборот удалено из липидного матрикса, оно может вызвать изменения в механических свойствах липидного бислоя и, следовательно, высвободить энергию, которая может быть достаточной для активации (или ингибирования) рецепторов клеточной поверхности в отсутствие их родственных лигандов. Истощение холестерина из плазматической мембраны может активировать рецептор эпидермального фактора роста (Chen, Resh, J. Biol. Chem. 2002, 277, 49631-49637) и Fas рецептор лиганд-независимым образом (Gniadecki, Biochem. Biophys. Res. Commun. 2004, 320, 165-169). Внутримозговое кровоизлияние вызывает лизис эритроцитов, утечку гемоглобина и его окисление в гемин. Подобно холестерину, гемин обладает жесткой структурой, и свободно проникает в бислой плазматической мембраны (Qutub et al., J. Phys. Chem. B 2010, 114, 4529-4535), что приводит к нарушению структуры микродоменов плазматической мембраны (рафтов). Это может налагать физиологически значимые эффекты на активность трансмембранных рецепторов и ионных каналов. Как показали наши модельные эксперименты на эритроцитах (Senchenkova E., Skvertchinskaya E., et. al. APS Conference, 2017, 20-4) гемин может служить триггером изменения формы клеток, и способствовать каскадированию образования гемина за счет высокой лизисной активности в отношении мембран эритроцитов. Пока нет доказательства прямого или опосредованного участия гемина в активации поверхностных рецепторов на нейроне, но гемин может, по крайней мере, способствовать индукции апоптоза или способствовать выживанию, при действии на нейрон других повреждающих стимулов. The structure and functions of the transmembrane proteins are critically dependent on properties of the surrounding lipid bilayer. Such properties of the lipid bilayer as thickness (depends on lengths of fatty acids), phase and fluidity (depends on temperature, degree of saturation of fatty acids, presence of cholesterol and sphingomyelin), curvature (depends on geometry of head groups and diacylglycerols composing phospholipids and propensity to formation of lamellar or hexagonal structures) influence activities of signaling proteins. If any chemical substance is embedded into or depleted from the lipid bilayer it can induce changes in mechanics of the lipid bilayer and release, therefore, energy which can be enough for activation (or inhibition) of the cell surface receptors in the absence of their cognate ligands. Depletion of cholesterol from the plasma membrane can activate epidermal growth factor receptor (Chen, Resh, J. Biol. Chem. 2002, 277, 49631-49637) and Fas (Gniadecki, Biochem. Biophys. Res. Commun. 2004, 320, 165-169.) in ligand-independent manner. Intracerebral hemorrhage induces lysis of erythrocytes, leakage of hemoglobin and release of hemin from hemoglobin. Damage to neurons produced by hemin is mainly due to oxidative stress because of iron bound to hemin. However highly lipophilic hemin can penetrate into the lipid bilayer of the plasma membrane (Qutub et al., J. Phys. Chem. B 2010, 114, 4529-4535) and doing so may impose physiologically meaningful effects on activities of the transmembrane receptors and ion channels. Similar to cholesterol hemin possesses a rigid structure. However highly lipophilic hemin can penetrate into the lipid bilayer of the plasma membrane (Qutub et al., J. Phys. Chem., 2010, 114, 4529-4535), which leads to a disruption in the structure of the microdomains of plasma membrane (rafts). This may impose physiologically significant effects on the activity of transmembrane receptors and ion channels. As demonstrated by our model experiments on erythrocytes (Senchenkova E., Skvertchinskaya E., et al. APS Conference, 2017, 20-4), hemin can serve as a trigger for changing the shape of cells, and facilitating the cascade of hemin formation due to high lysis activity against erythrocyte membranes. Even if it is not likely that hemin may activate surface receptors on the neuron, hemin may, at least, facilitate induction of apoptosis, or promote survival when neuron is challenged with other noxious stimuli.
