Today, some of the existing animal species that live on our planet are threatened with extinction. New opportunities for the conservation of the genetic biodiversity of the animal world are provided by modern biotechnologies based on the achievements of reproductive biology. It is about cryopreservation of reproductive material of valuable sires. One of the most important tasks in preserving genetic material is the creation of new methods and media for freezing and defreezing the genetic material of males of different species and breeds. This is possible through the improvement of freezedefreeze media that contribute to the conservation of genetic information and protection the morphological, physiological, biochemical parameters of the reproductive material of males in the cryiopresevation. The aim of creating cryoprotective media is to stabilize the functional activity of biological objects after their storage at extremely low temperatures and to stabilize morphological parameters, in particular, to reduce gametopathologies in the reproductive material, investigated the effectiveness of various carbohydrates in synthetic media for diluting and freezing the reproductive material of studbulls. The process of cooling and preparing reproductive material for freezing at extremely low temperatures is accompanied by destruction associated with thermal and osmotic stress. As a result of such disorders, the vital components of gametes can enter the intercellular space. Incidentally, a destruction of architectonics can be associated with complex biochemical, biophysical and functional changes in the cells. According to the obtained experimental data, in the study of 2000 gametal cells (90.4 ± 1.31 % - normal and 9.6 ± 1.31 % - with pathologies), in the case of using glucose-glycerol-vitelline medium, 74.8 ± 1.50 % of the spermatozoa proved to be with normal architectonics, and 15.2 ± 1.50 % - with pathological changes, and when using media based on lactose and raffinose, the number of pathological forms increased to 19.4 ± 1.77 % in the case of lactose and 36.2 ± 2.14 % in case of raffinose. This indicates the role of molecular mass in osmosis and the metabolism of nutrients necessary to maintain the life activity of gametal cells in the process of freezing-defreezing. |
