Rezumat

Propose: to study the allelic polymorphism of genes encoding the enzymes of estrogen metabolism in elderly people and long-livers of Belarus. Two study groups were formed: a group of long-livers (over 90 years of age) consisting of 136 people and a group of elderly people comprising of 67 people (42 women and 25 men). Isolation of total DNA was carried out using the method of phenol-chloroform extraction and ethanol purification. CYP19A1 (rs10046, rs4646) and СOMT (rs4680 and rs4818) genotyping was performed using PCR followed by sequencing. The content of total testosterone, estradiol, and 17-OH progesterone in blood serum was determined in patients. The statistical analysis was carried out using the GraphPad InStat Version 3.05 application software. Analysis of a degree of incidence of the genotypes of the investigated polymorphisms of the CYP19A1 and COMT genes when comparing the group of long-livers and the control did not demonstrate statistically significant differences. Further, we analyzed the distribution of polymorphic variants of CYP19A1 and COMT genes depending on the body mass index (BMI) in the investigated groups. It was shown that the carriers of the CT genotype of the rs10046 polymorphism of the CYP19A1 gene are more frequently occurring among the people aged 60-89 years with an increased BMI (69.0%) compared with the long-livers with an increased BMI (30.4%) (OR = 0.22; CI: 0.06-0.80; p = 0.018). This may indicate the influence of CYP19A1 polymorphism on the tendency to overweight. Both overweight and obesity constitute a risk factor of the development of chronic diseases associated with metabolic disorders and lead to early mortality among the population. No associations were found between the polymorphic variants of the CYP19A1 and COMT genes and the mean values of testosterone, estradiol, and 17-OH progesterone in blood serum. The results obtained may be used to improve the prevention and diagnosis of diseases associated with metabolic disorders in the groups of elderly people.
