Rezumat | ||||||
Background. Due to lack of cellular immunogenic components, decellularized natural scaffolds are considered an optimal solution in development of tissue-engineered vascular grafts. Objective of the study. The goal of this study was to evaluate the effect of commonly used detergents in vascular tissue decellularization and to understand their potential impact on structural changes. Material and Methods. Fresh porcine aortas (lenght 60–160mm, lumen diameter 15–23mm, wall thickness 2–3mm) were obtained from a local slaughterhouse. The samples were cleaned off excess connective tissue and fats and rinsed in dH2O for 24h. Results. The research protocol included 6 groups, including one control group and 5 experimental: a. 1%SDS; b. 1%SDC; c. 1%Triton X-100(TX); d. 0,5(SDC+SDC)%; e. 0,5(SDS+SDC+TX)%. In each case 0,02%EDTA were added. The samples were subjected to continuous shaking at 37oC for 48hrs (n=3 for each group). Then, the vessels were rinsed 24hrs in PBS and stored in saline solution 4oC for further examinations. Samples from obtained segments were stained with H&E. The thickness of the intima and media were also measured. In addition, remnant DNA were quantified with the GeneJET Genomic DNA Purification Kit. Conclusion. Triton X-100 based decellularization technique were found to be more effectively in cellular components elimination. However, biocompatibility and mechanical properties assessments should be carried out in future studies |
