Optimal proliferation of callus from Rhodiola rosea L. on MS solid and liquid medium occurs when the ratio between BA and ANA is maintained at 1:2. Addition of Kinetin in this medium does not support biomass growth. The growth of callus represents a sigmoid curve with three distinct phases: lag phase (from 0 up to 30 days), exponential (30-50 days), and stationary (50-90 days). Initiation of stationary phase can be marked by the polypeptide spectrum analysis, which becomes richer. The end of stationary phase is determined by degradation of polypeptide with relatively high molecular weight. |
