The main goal of this work was to determine the influence of meteorological conditions on the spread of phytoplasma in the tomato field. Molecular diagnosis was carried out on plants of two tomato varieties during three growing seasons: 2016, 2017, 2018. DNA for nested PCR analysis was isolated from the peduncles of tomato using the alkaline express method. Probes (peduncles) for analysis were collected in the middle of July, August and September. It was established that the distribution of phytoplasma infection was nonuniform in the two analyzed varieties across the three seasons of vegetation. Climatic conditions of the summer of 2016 and 2018 were relatively similar characterized by high temperatures, especially in August. The summer of 2017 was cooler and wetter. Consequently, the percentage of infected tomato plants was significantly lower, only about 42%, at the end of the period of vegetation in 2017 versus 92% in 2016 and 70% in 2018. We can suggest that lower temperatures reduced the activity and reproduction of insect vectors of phytoplasma. As a result, the percentage of infected plants in tomato field did not essentially increase. The negative effect of precipitations on the spread of phytoplasma was not established. Nevertheless, it is necessary to take into account combinations of different climatic factors as well as some specific characteristics of tomato varieties when estimating the risk of the infection spread. |
