Rezumat

Conventional PCR is generally considered a qualitative method. But using serial dilution and partitioning of reaction mix with the following application of statistical analysis permits to use conventional PCR as a quantitative or semi-quantitative method. Samples of seed material of maize ‘CP 137’ and ‘CP 148’ genotypes were analyzed for accumulation of toxigenic A. flavus strains. Partitioning and Poisson statistics were applied as an alternative quantitative approach for conventional PCR. ‘CP 148’ maize seed samples revealed a significantly lower quantity of aflatoxin-producing Aspergillus DNA.
