Derivaţii naftochinonei sunt cunsocuţi pentru activitatea lor anticanceroasă, antituberculoasă, antifungică şi antibacteriană. Scopul principal al cercetărilor prezente a fost obţinerea derivaţilor noi de 1,4-naftochinonă cu solubilitate crescută în apă şi farmacocinetica dirijată în organismul uman. Scopul propus a fost atins atât prin introducerea grupărilor hidrofile în structura 1,4-naftochinonei, cât şi prin introducerea unui fragment de structură sensibil la anumite esteraze. Întai, juglona a fost obţinută din 1,5-dihidroxinaftalină prin oxidarea catalitică în acetonitril cu randamentul 70% în concordanţă cu metoda descrisă anterior de către Andrusenco în 2017. Apoi, juglona a fost gradual transformată în 2-hidroxijuglonă, derivatul ei cu N-acetil-L-cisteină şi, în final, în derivatul dipeptidic cu esterul ciclopentilic al L-leucinei. Produsul intermediar al 2-hidroxijuglonei şi N-acetil-L-cisteinei a fost obţinut cu un randament mai mare decât cel publicat în literatura de specialitate de către Sabutskii et al. Structura derivatului dipeptidic a fost confirmată prin analiza RMN. În concluzie, s-a constatat că compusul ţintă a fost obţinut printr-o cale eficientă şi cu randamentul şi puritatea satisfăcătoare.
Naphthoquinone derivatives are known for their anticancer, antituberculosis, antifungal, and antibacterial activity. The main objective of the present research has been obtaining of the new derivatives of 1,4-naphthoquinone with higher solubility in water and designed pharmacokinetics in the human body. The objective was reached through introduction of hydrophilic groups in the structure of 1,4-naphthoquinone, as well as addition of a motif with selective sensitivity towards certain esterases. First, juglone has been obtained from 1,5-dihydroxynapthaline by catalytic oxidation in acetonitrile with a 70% yield according to the method previously described by Andrusenco in 2017. Then, juglone was gradually transformed into 2-hydroxyjuglone, its derivative with N-acetyl-L-cysteine and, finaly, into the dipeptide derivative with cyclopentyl ester of L-leucine. The intermediate product of 2-hydroxyjuglone and N-acetyl-L-cysteine has been obtained with higher yield than that published in the specialized literature by Sabutskii et al. The obtained dipeptide derivative structure has been confirmed by NMR analysis. It has been concluded that the desired compound has been obtained via an efficient route and with satisfactory yields and purity. Bibliografie/ Bibliography 1. Andrusenco T.
