Teza

Protein C and X factor are the most important proteases of hemostatic system. Protein C plays role in regulation of anticoagulation, inflammation, cell death, and maintaining the permeability of blood vessel walls in humans. Activated protein C performs these operations primarily by proteolytically inactivating of proteins factor Va and factor VIIIa. Protein C deficiency is a congenital or acquired (liver diseases, disseminated intravascular coagulation, sepsis, indirect anticoagulants) condition that leads to increase of risk for thrombosis. Deficiency of factor X may present with epistaxis, hemarthrosis and gastrointestinal blood loss. There are also both inborn deficiency Х factor and acquired (liver diseases, disseminated intravascular coagulation, sepsis, amyloidosis, uremia, indirect anticoagulants). Personalized medicine is a very popular today. In this model, diagnostic testing is often employed for selecting appropriate and optimal therapies based on the context of a patient’s genetic content or other molecular or cellular analysis. Some of the proteins in snake venom have very specific effects on protein C and X factor. But this diagnostic system is expensive. It was discovered, that Aspergillus ochraceus proteases have a direct influence on protein C and coagulation factor X. It is perspective area in development of available and qualitative diagnostic system. The aim of this work was selection of optimum concentrations range of lyophilized preparation and isolated activators by isoelectric focusing of extracellular protease-activator of micromycetes Aspergillus ochraceus L-1. These proteinases have protein C and factor X activator activity. It was demonstrated that specific activity with protein C and factor X was 33,4 unit/ml ×10 -3 and 18,6 unit/ml ×10 -3. Optimal range of lyophilized extracellular preparation of microscopic fungus Aspergillus ochraceus L-1 was 0,5–1,0 μg / ml for activated protein C and 0,5 – 1,75 μg / ml for factor X. For proteins which were isolated from cultural medium of Aspergillus ochraceus L-1 recommended active concentration for activated protein C was 0,5–1,0 μg / ml and for X factor was 2,0–3,25 μg / ml. To sum up, proteinases of micromycetes produced by Aspergillus ochraceus L-1 is very perspective for protein C and X factor diagnostics. This method may be is cheaper and easier than another methodics.
