Rezumat

At the Fruit Research Station of Cluj research was done regarding in vitro propagation and ex vitro acclimation of Sequoia sempervirens. The highest multiplication rates were obtained on hormone-free WPM medium, but the plantlets cultured on hormone-free MS medium were more vigorous. The presence of BAP in the culture medium resulted in the regeneration of short shoots, callus and masses of buds. For the in vitro rooting of Sequoia sempervirens 5 variants of media were tested, resulting poor rooting percentages of up to 25 %. The variants of media containing 0.5 and 1 mg/l IBA inhibited the rooting of Sequoia plantlets and caused the abundant generation of callus at the base of the explants. The rooted plantlets were successfully acclimated in hydroculture and the acclimated plants were further grown in containers with a potting mix of soil and peat.
