Background: Pathogenesis in recurrent pregnancy loss (RLP) is complex and involves the interaction of multiple genetic and environmental factors. Is supposed that genetic polymorphisms in MTHFR (C677T and A1298C), MTR (A2756G) and MTRR (G66A) genes may be associated with the risk of recurrent pregnancy loss. In this study, we aimed to investigate the association between the polymorphism of MTHFR, MTR, MTRR genes and the pregnancy loss. Methods: In this study we determined the genotypic frequencies of the MTHFR, MTR, MTRR gene polymorphisms in a sample of 274 women from Moldova, 166 with 2 and more miscarriages and 108 without history of miscarriages, with 2 healthy births. The genomic DNA was extracted from the peripheral blood collected on EDTA. The methods used to study polymorphisms were PCR and PCR-RFLP. Statistical analysis was performed using www.gen-exp.ru site. Results: The results of this study showed that women with recurrent pregnancy loss have a higher proportion of the MTHFR677TT, MTHFR 677CT, MTR2756AG, MTRR66GG genotype compared with the control group, does not present significant differences between the study group and control (p>0.05). The test Hardy-Weinberg was applied for the control and research group. Have been established no statistically significant differences (p>0.05) between the observed and theoretically expected genotypic frequencies for MTHFRC677T, MTHFR A1298C, MTR A2756G polymorphisms, but for MTRR G66A polymorphism have found the Hardy-Weinberg disequilibrium in control and research groups (χ2=22.81, p=0.000, respectively χ2=28.31, p=0.000). Conclusion: Our study revealed the role of folate and methionine cycle genes polymorphisms (MTHFR, MTR, MTRR) in the etiology of recurrent pregnancy loss showed no correlation between the results obtained from the two groups (χ2<5, p>0.05), but the risk of pregnancy loss is higher for women carrying MTR 2756AG genotype, OR=1.38, 95% CI [0.81-2.34], as well as MTRR 66AA genotype (OR=1.19, 95% CI [0.21-6.59]). |
