The genetic polymorphism of beta-1-adrenoreceptors and cytochrome P-450 enzymes may determine the efficacy and harmlessness of beta-adrenoblockers in the treatment of cardiovascular disease (hypertension, ischemic heart disease, arrhythmias and heart failure). The genetic polymorphism of the ADRB1 and GRK4 genes, through G protein-coupled receptors, determines the response to drugs by influencing beta1-adrenergic receptor-mediated intracellular signaling. Most isoenzymes (CYP2D6, CYP3C9, CYP3C19, CYP3A4) involved in the metabolism of beta- adrenoblockers have genetic polymorphism, which may influence the results of treatment and the probability of adverse reactions depending on cardiovascular disease and concomitant diseases. Most isoenzymes involved in the metabolism of beta-adrenoblockers have genetic variations, which may determine the efficacy and harmlessness of these drugs. For these reasons, the study of pharmacogenetic peculiarities in the patient is considered the primary tool in the realization of personalized medicine. |
