Rezumat

The applied part of cryobiology – cryoconservation, was become a current biotechnological method of cryopreservation of genetic resources in the form of sperm collections and has been successfully applied in cattle breeding. Nevertheless, 50% of semen sells do not restore their biological value after thawing. The purpose of our investigation – evidence of cryogenics response of semen proteins, bloods lipids metabolism and the homoeostasis of the male sexual hormones in sire bulls and the influence of biopreparation – BioR, obtained from Spirulina platensis on cryobiological indices of bull semen material. The investigation has been performed on bulls from the Holstein and Black Spotted breed of Moldavian type, maintained during the investigation in adequate conditions from the point of view of microclimate and fodder. The bioextract administrations have been done daily during 10 days in volume of 0,2 ml/100 kg living mass/day. Having as purpose the research of cryoconsevation influence on the bill semen proteomics and the effect of BioR administration on the biochemical and the cryobiological semen indices, in the control and experimental groups have been studied: the mobility, the longevity and the survival absolutly indices of the thawing spermatozoids; some metabolics tests: such as proteins content in the raw, and thawed semen, the concentration of blood cholesterol, DMA and circulating testosterone concentration after BioR administration. The studded index was determined utilizing the preparation produced by “Lyonorm” and “La Chemia” (Brno), “Biocont” (Russia). Indices of semen quality were determined by general accepted method in the laboratory for semen microscopic examine. Results of our experiments concerning the content of total proteins in the raw and thawing bull semen material denote no difference between the value of this parameter in the fresh and cryopreserved-thawed bull gametes. By the same token, the structural gametes proteins posed the resistance given the influence of ultra low temperature (-196°C). The study on the raw gametes the rate of some proteins fractions in accordance with their isoelectric point and the consequents of the thermal stress determinates by cryoconservation process indicate that both in the raw and thawed semen cells the most rate occupy the hydrophilic proteins. The lowest rate owns the basophilic fraction. The acidophilic proteins occupy a middle position in this rate hierarchy. Similar to totals proteins, after semen conservation-thawing process neither of theirs support the significantly modifications. It was observed a tendency of the diminution of the hydrophilic proteins (- 3,35%) and an increase of the basophilic proteins (+ 2,78 %). The analysis of the studded proteins fractions has evaluated that in the raw gametes prevailed γ-globulins. Conservation and thawing process of the semen material was associated by an increase of the albumins rate. The rate of other three proteomic fractions: α-, β- and γ-globulins was decreased given to theirs value registered in raw gametes. The influence of the intramuscular administration of BioR preparation during 10 days in the sire bulls on the albumins, α-, β- and γ-globulins fractions content certify any modification of the studded proteomic fractions rate in thawed bull semen cells, given the control (semen cells of non treated by BioR bulls). The γ-globulins rate was increased with 19,28% in the experimental group. Also it was observed an increase of β- and α-globulins by 8,5% and 2,36%, respectively, given the control group. In the same condition the value of albumins rate, in the thawed semen cells of experimental bulls was decreased with 32,86% given the non treated by BioR preparation. The force of circulating lipids peroxidation in the harvesting blood samples from bulls denote that the DMA concentration in the blood samples after administration of bioextract decreased with 16,58% given the experimental group. Dosing the DMA concentration in the blood samples drayed 50 days after ceasing the administration of biopreparation denote that the lipids peroxidation has been continued to decrease with a difference of 5,45% comparative with results registered after ceasing of biopreparation administration and with 21,12% given the preexperimental period. The promising modifications where established given dynamic of blood cholesterol concentration after administration of biopreparation. It was was certified a meaningful decrease of cholesterol concentration on the experimental group with 24,30%. Thus the preparation BioR, modulate different the blood lipids fractions taken in evidence, manifesting the anabolic effect given triglycerides, ß-lipoproteins and the catabolic effect concerning the cholesterol. Because the male sexually hormones are of steroid origin the decrease of blood cholesterol was associated by the testicular steroid genesis, expressed through increase of concentration of circulating testosterone in the experimental sire bulls. with 150%.The highest result was inscribed counterpart the survival absolutly indice and the mobility of the thawing bull spermatozoids. In the case of the longevity it was established a temperate increase. In acordance with the established results of modifications of the proteomic fractions rate in semen cells proteomic fractions decrease rate in semen cells and cryobiological parameters of thawed bull semen, it is posible to mention that biopreparastion BioR have been atenuate the cryogenics response of some semen cells proteomic fractions and the speed of reaction of lipids peroxidation and DMA contentration, respectively increasing the cryobiological characteristics of the thawed bull semen. Since thus parameters are a high influence on the spermatozoids skill fecundity it can to conclude that the BioR will increase the rate of the fecundity of famales after fist artificial insemination. Structural gametes proteins posed the resistance given the influence of ultra low temperature (-196°C), content of total proteins in bull semen material denote no difference between value of this parameter in the raw and cryopreserved-thawed bull gametes. It inhibit the chain reaction of frees radicals of blood lipids peroxidation and thus reduce the blood quantity of DMA, reducing considerably the amount of circulating cholesterol; have an stimulatory effect on the hypothalamus-hypophysis-testicle system for the testosterone secretion.The BioR has an evident influence on the cryobiological specifics features of spermatozoids, such as the seminal cells mobility, in the same condition, the longevity and the survival absolutly indice are intensely influenced. A comparative analysis of these tables shows that families of the Carpathian breed (with queens of the second and third year of breeding) were better in their development than the Ukrainian steppe bees by an average of 3.2% in 2014 in the apiaries of the Mykolaiv region. Comparing the laying of eggs by uteruses of the second and third year of hatching in apiaries of Odessa and Vinnytsia regions, it should be noted that the uterus of the Ukrainian steppe breed in apiaries of Odessa region laid more eggs than the uterus of the Carpathian breed in 2014 by 8.3% and the first apiaries, 7% in terms of second apiaries. Also in terms of egg-laying, the uterus of the Carpathian breed was inferior to the Ukrainian steppe uteri, which began to gain a high rate of egg-laying and exceeded the Carpathian ones. This circumstance gives a number of advantages to Carpathian bees over Ukrainian queens, in better use of early honeybees or opportunities for beekeepers to form earlier layers from them. Thus, Ukrainian steppe bees emerged more moderately from the wintering state, so they showed their maximum rate of development a little later. This behavior of these bees can be used for later honey collection, when families are gaining enough strength and can use it well.
