Background. Benzodiazepines are a class of drugs known as minor tranquilizers. They are used in : anxiety disorders, seizures, insomnia. Inappropriate use and the irresponsibility of specialists has led to the appearance of serious, obvious side effects. Objective of the study. The aim of the study is to emphasise benzodiazepine`s metabolism and establish possible interactions. To identify the beznodiazepines with the lowest probability of adverse reactions in specific conditions. Material and Methods. Evaluation and processing the data present in the bibliographic sources for 17 studied benzodiazepines. Number of bibliographic sources on the studied compartments: general characteristic (20), pharmacokinetics of benzodiazepines (30), mechanism of metabolic action (50). Results. Benzodiazepines have become some of the most widely used minor tranquilizers. CYP450 cytochrome enzymes, located mainly in the liver, are responsible for their metabolism. Taken in optimal doses, as monotherapy, benzodiazepines causes low toxicity. Short-acting compounds are more suitable for the elderly, and those with long-acting action may accumulate over time. Short-acting compounds are more prone to addiction. In a short period of time, can be used benzodiazepines with an intermediate half-life, as the risk of adverse reactions is low. Conclusion. Knowledge of the metabolic mechanism of benzodiazepines, metabolizing enzymes, inhibitors and inducers is important for avoiding adverse reactions. |
