DNA from different tomato cultivars was screened for the presence of the Mi gene that confers resistance to the root-knot nematodes. A codominant PCR marker REX-1, closely link to Mi, was used in this analysis. PCR of REX-1 resulted in an amplification product of 750 bp in all analyzed tomato cultivars. After digestion of amplicons with TaqI, one genotype (Djina)yielded three fragments - 750, 570 and 160 bp - was identified as heterozygous for Mi resistance gene (Mi/mi). Such screening is a preliminary study and further investigation will permit the identification of resistant cultivars and their use in breeding programs. |
