Rezumat

Active organometallic molecule inhibitors CMT-67, CMSA-4 were tested for their ability to induce apoptosis of еpithelioid cervix carcinoma cells of line HeLa. Viability of the cancer cells was determined by Alamar Blue assay using oxidation-reduction indicator - Resazurin. In order to detect the eventual presence of concomitant adverse effects associated with the prooxidant properties of experimental coordination compounds, was used method of quantitating the oxygen radical absorbing capacity (ORAC), which was adapted to reader (Synergy, BioTek). Antiproliferative activity experiments determined, that the IC50 values are 2, 1±5 μM/L for CMT-67; 3±0,15μM/L for CMSA-4 and 6±0.1μM/L for Doxorubicin (DOX). Thus was established that, compounds CMT-67 and CMSA-4 exhibit stronger inhibitory activity on HeLa cell line proliferation than DOX, so the half maximal inhibitory concentration for CMT-67 is 3 times less and CMSA-4 is 2 times less than DOX. Analyzing the ORAC results, we observe that two compounds showed the best antioxidant activity (AOA) compared with Trolox and DOX. Kinetic curves of Trolox demonstrate concentration dependent protection of fluorescein against oxidative degradation by AAPH (Figure 1). The AUC and the Net AUC were calculated using equations. Antioxidant property was determined by the ratio of the slope (m) of the linear regression curve. Slope values are 3,5 for CMT-67; 2,18 for CMSA-4; 0,5 for DOX and 1,2 for Trolox. The calculated Trolox equivalents were used for comparative analysis of the AOA. The highest antioxidant activity is shown - CMT-67. Correlation between the antiproliferative activity on HeLa cells and AOA of substances CMT-67, CMSA-4 and DOX was calculated (R2-0,98), which is statistically significant evidence of strong correlation between the antiproliferative and AOA. Is very important that the anticancer substance due to their antioxidant potential may exert effects by interfering with oxygen free radicals, which act as second messengers in the critical signaling pathways for cancer cells proliferation. Therefore, the complexes CMT-67, CMSA-4 are of interest for further study for subsequent use their as antitumor agents.
