Нейродегенеративные заболевания (НДЗ) являются одной из главных проблем современной неврологии. Для подавляющего большинства НДЗ характерен длительный латентный период развития и прогрессирующее течение, приводящее к выраженной инвалидизации и смерти пациентов. Болезнь Паркинсона (БП) одно из наиболее распространенных нейродегенеративных заболеваний. Болезнь сопровождается поражением структур экстрапирамидной моторной системы, при этом происходит прогрессирующее разрушение и гибель дофаминергических нейронов. Основная проблема в изучении всех НДЗ состоит в невозможности получения биоматериала в процессе развития заболевания, и, следовательно, невозможности изучения молекулярно-генетических изменений приводящих к нейродегенерации. Для решения этих задач в настоящее время активно используют индуцированные плюрипотентные стволовые клетки (ИПСК) человека. ИПСК, полученные от пациентов с наследственными формами НДЗ, несут соответствующие мутации во всех своих производных и, как уже показано многими авторами, демонстрируют патологический фенотип, присущий определенному НДЗ. В нашей работе была исследована экспрессия ряда генов, кодирующих нейротрофические факторы в первичных фибробластах и в нейрональных клетках ( полученных из ИПСК) разной степени зрелости, от пациентов с спорадической и наследственными формами болезни Паркинсона, а также здоровых доноров. Было показано, что уже в исходных фибробластах у пациентов с БП снижен уровень экспрессии мРНК гена BDNF. Нейрональные клетки, находящиеся на разных стадиях дифференцировки, с мутацией в гене LRRK2 демонстрируют увеличение экспрессии, тогда как клетки с мутацией в гене PARK2 напротив – снижение экспрессии большинства исследуемых генов нейротрофических факторов. Neurodegenerative diseases are one of the main problems of modern neurology. For the vast majority of neurodegenerative diseases, there is a long latent period of development and a progressive course leading to severe disability and death of patients. Parkinson's disease (PD) is one of the most common neurodegenerative diseases. The disease is accompanied by damage to the structures of the extrapyramidal motor system, with the progressive destruction and death of dopaminergic neurons. The main problem in the study of all neurodegenerative diseases is the impossibility of obtaining a biomaterial in the course of the development of the disease, and, consequently, the impossibility of studying the molecular-genetic changes leading to neurodegeneration. To solve these problems, currently induced human pluripotent stem cells (IPSC) are actively used. IPSC obtained from patients with hereditary forms of neurodegenerative diseases carry the corresponding mutations in all their derivatives and, as already shown by many authors, demonstrate a pathological phenotype inherent in a specific neurodegenerative diseases. In our work, expression of a number of genes encoding neurotrophic factors in primary fibroblasts and in neuronal cells (derived from IPSC) of varying degrees of maturity was investigated, from patients with sporadic and hereditary forms of Parkinson's disease, as well as healthy donors. It was shown that already in the fibroblasts in patients with PD, the level of mRNA expression of the BDNF gene was reduced. Neuronal cells at different stages of differentiation with a mutation in the LRRK2 gene show an increase in expression, whereas cells with a mutation in the PARK2 gene, in contrast, decrease the expression of most of the studied genes of neurotrophic factors.
