Окислительный стресс является одной из причин повреждения и гибели нервных клеток при ишемических поражениях мозга. Инсулин обладает нейропротекторным эффектом, но механизм его действия далек от понимания. Целью нашей работы является изучение влияния инсулина, вводимого интраназально крысам, подвергнутым ишемии и реперфузии мозга (ИР), на накопление продуктов перекисного окисления липидов (ПОЛ) и активность суперооксиддисмутазы (СОД). Двухсосудистую ишемию переднего мозга вызывали у крыс линии Вистар весом 270-330 г путем окклюзии каротидных артерий на 20 мин в сочетании с гипотензией. Затем проводили реперфузию мозга в течение 1 ч. Инсулин в дозе 0.5 IU вводили крысам интраназально за 1 ч до окклюзии. Далее определяли содержание диеновых и триеновых конъюгатов (ДК и ТК), продуктов ПОЛ, в липидном экстракте и активность СОД в митохондриальной фракции коры мозга крыс. В коре мозга крыс, подвергнутых ИР, происходило достоверное увеличение уровня ДК (76±6.2 у.е./мг липидов) и ТК (17.9±1.55 у.е./мг липидов) по сравнению с контрольной группой (59.8±5.5 и 14.1±0.55у.е./мг липидов, соответственно), интраназальное введение 0.5 IU инсулина снижало содержание ДК и ТК до уровня контрольных значений. ИР сопровождалась увеличением активности СОД (0.091±0.013 у.е./мг белка) по сравнению с контрольной группой (0.05±0.006 у.е./мг белка), введение 0.5 IU инсулина привело к достоверному снижению активности СОД (0.02±0.005 у.е./мг белка) в митохондриальной фракции коры мозга крыс. Таким образом, инсулин при интраназальном введении способствует уменьшению развития окислительного стресса при двухсосудистой ишемии и реперфузии переднего мозга крыс. Oxidative stress is one of the main causes of damage and death of neurons in the ischemic brain. Insulin is known to be neuroprotective, but the mechanism of its action is far from being understood. The aim of our work was to study the effect of insulin intranasal administration to rats subjected to ischemia and reperfusion of the brain (IR), on the accumulation of lipid peroxidation (LPO) products and the activity of superoxide dismutase (SOD). Two-vessel ischemia of the anterior brain was caused in the Wistar rats (270-330 g) by occlusion of the carotid arteries for 20 minutes in combination with hypotension. After the blood flow recovery the reperfusion of the brain was maintained within 1 h. Insulin were administered intranasally to rats 1 hour before occlusion at the dose of 0.5 IU. The content of diene and triene conjugates (DC and TC), products of LPO, in the brain lipid extracts and the activity of SOD in the mitochondrial fraction of rat cerebral cortex were determined. In the cortex of rats subjected to IR, the level of DC (76±6.2.e./mg of lipids) and TC (17.9±1.55 c.u./mg of lipids) have significantly increased in comparison with the control group (59.8±5.5 and 14.1±0.55 c.u./mg lipids, respectively), intranasal treatment of the insulin at the dose of 0.5 IU reduced the content of DC and TC to the control values. IR was accompanied by the increase of the SOD activity (0.091±0.013 c.u./mg protein) compared with the control group (0.05±0.006 c.u./mg of protein), intranasal injection of 0.5 IU insulin resulted in the significant decrease of the SOD activity (0.02±0.005 c.u./mg protein) in mitochondrial fraction of rat cerebral cortex. Thus, insulin, administered intranasally at the dose of 0.5 IU, attenuates the development of oxidative stress under conditions of the two-vessel ischemia and reperfusion of the rat brain.
